Using Microscopy is a series of four 15-20 minute video programs highlighting the relevance and impact of modern microscopy technology and methods on our understanding of life's diversity. Program 1 demonstrates proper setup and adjustment of the light microscope to achieve images of the highest possible quality. Key properties of light and lenses--magnification, diffraction, refraction and reflection, resolution and contrast--are explained in the context of the various adjustments. The second program in the series introduces three powerful contrast-generating modes of microscopy. It covers phase contrast, differential interference contrast (DIC), and immunofluorescence microscopy, and points out the advantages of each. The third program introduces a type of microscopy with yet a higher resolving power, electron microscopy. Included is a discussion of the two most common forms of electron microscopy--transmission and scanning EM--in the context of an ongoing infectious disease investigation. Finally, Program Four in the series demonstrates how a functional cell component can be traced in living cells using a fluorescent analog and special fluorescence imaging techniques.
